Rooster sperm pellet cryopreservation protocols: effect of step variations on the qualitative parameters of post-thawed sperm

The cryopreservation of sperm into pellets is not the preferred way to package avian semen but quick and easy do does require sophisticated technology. aim this study was evaluate rooster viability mobility incidence normal cells injuries in post-thawed pelleted sperm. outcomes different pelleting protocols were evaluated, which varied according parameter combinations used each critical steps freezing process thawing conditions differed methodology temperature. employing 6% DMA showed highest values thawed mobility. most favourable method terms using hot-plate at 60 °C, followed by water-bath 50 °C. resulting best quality parameters employed a 1:2 dilution rate, 30-min equilibration time 4 DMA, 80 µL °C or According highly susceptible damage caused freezing-thawing methodology, although survival rate still reached 39%, with 32% recovered respect fresh samples. HIGHLIGHTSA feasible for ex-situ management genetic resources birds involves use cryopreserved sperm.The domestic fowl constitutes reasonably simple, affordable technology.This technology may be extremely valid practical on small farms.
